Research Abstract

Spatio-temporal patterns of excitation propagation were recorded in the rat auditory cortex in slice preparations (400 mum thicknesses) by optical recordings using voltage-sensitive dyes. For an absorption type dye, slice preparations were illuminated by a light passed through an interference filter (700 * 30 nm). For a fluorescence type dye, slice preparations were excited by a light passed through an interference filter (520 * 45 nm), the emitted fluorescent light was transmitted by dichroic mirror (575 nm) and collected through a long pass filter (>610 nm). The image of the slice was focused on a 128 x 128 pixel camera and optical responses elicited by stimulation were recorded every 0.6 ms.
Electrical stimulation of the border between the white matter and layer VI evoked vertical as well as horizontal spreading responses. The horizontal propagation was widespread and strong especially in layers II/III in auditory cortex slices. The extent of the horizontal excitation in the auditory … More cortex was approximately double those reported in the visual cortex. Electrical stimulation of supragranular layers elicited widely propagating horizontal excitation along supragranular layers with little horizontal excitation in infragranular layers. A horizontal cut along layers was made between supra- and infragranular layers to examine whether interactions between supra- and infragranular layers are necessary for a horizontal propagation of excitation. Electrical stimulation of supragranular layers elicited horizontally propagating excitation in supragranular layers with the same propagating velocity as that seen before a cut was made. Electrical stimulation of the infragranular layers elicited horizontally propagating excitation in the infragranular layers with the same propagating velocity as that seen before a cut was made, although excitation in supragranular layers disappeared.
These results suggest that the horizontal propagation of excitation in supra- and infragranular layers are widespread and can occur independently in the auditory cortex. Less
