Research Abstract |
1. The implantation of ovarian tissue into the splenic capsule of gonadectomized female rats developed into benign granulosa cell tumors between 6, 8, and 10 months after autotransplantation. We investigated the immunohistochemistry of inhibin, activin, and follistatin in the implanted ovary, and the serial serum assay of inhibin, luteinizing hormone(LH), and follicle-stimulating hormone(FSH). The serial serum hormone assay revealed that the concentration of serum inhibin and FSH had an inverse relationship. The serum FSH level raised rapidly after implantation and this high level was sustained for at least 120 days after operation. Then, the marked increase of inhibin concentration was accompanied by a sudden decrease of FSH concentration. In contrast, LH level fluctuated in all cases. In immunostaining of inhibin, a high percentage of the tumor cells revealed an intensive fine to coarse granular positive reaction in the cytoplasm. In contrast to inhibin, activin and follistatin immunostaining was not identified in tumor cells. In conclusion, granulosa cell tumors secreted inhibin, and serum inhibin level elevated. Inhibin can be used as a histopathological and serological marked of granulosa cell tumors. 2. In hypogonadal mouse(hpg mouse), the implantation of ovarian tissue into the splenic capsule of gonadectomized female mice could not develop into any tumors. Further investigation should be required, but it would be difficult to develop granulosa cell tumor in hpg mice. 3. In domestic animals, mainly in dogs, the immunostaining of inhibin revealed various positive reactions in the granulosa cell tumors. Inhibin can be used as a histopathological marked for granulosa cell tumors.
