Research Abstract

分子構造から見るとプロスタグランジンは、プロスタン酸を基本骨格とし、五員環構造と側鎖の二重結合数によって分類される。即ち、五員環構造よりD,E,F,Iの4タイプとオキサン環を持つTXに分類される。各プロスタノイドが非常に似通つた構造を持つにもかかわらず、各プロスタノイド受容体は全体としてはその相同性が低い。しかし、プロスタノイド受容体はロドプシン型受容体ファミリーに属し、部分的にはプロスタノイド受容体サプファミリーとしての共通構造を持っている。
本研究は、DP/IPキメラ受容体を用いた解析を行い、リガンド結合親和性を比較することにより、リガンドの特異的結合を決定する受容体領域の同定を試みた。その結果、初年度は受容体の第2〜3膜貫通領域がプロスタノイド分子の5員環を認識し、第6〜第7膜貫通領域が側鎖の二重結合の有無を識別することを明らかにした。次年度は、さらに細かく部位を限定したキメラ受容体を用いて解析を進めた。その結果、受容体の第2膜貫通領域が5員環の認識に重要であることを解明した。ついで、この部位でIP受容体とDP容体間で異なっている4個のアミノ酸に点変異を加えた受容体を作成し、解析を進めた。その結果、5員環の認識に重要な役割を果たすアミノ酸の同定に成功した。
