Research Abstract |
A novel method for labeling of sugar chain, which can be used for analysis of sugar chain by HPLC, ELISA and TLC immunoblotting was investigated for simplification of analysis of sugar chains. To prepare reagents which have sufficient hydrophobicity to bind to solid-phases, ELISA plates or blotting membranes, aromatic aminoester compounds (amino-C&46) were synthesized from aminobenzoic acid and aliphatic alcohols (C8-C 16). Analysis of ABO blood group active sugar chains obtained from saliva mucin was performe4 by using the newly developed labeling reagents. However, we could not obtain satisfactory results. Although we could not obtain novel labeling reagents which were analyzable by HPLC and have sufficient hydrophobicity to bind solidphases for immunological analyses, we have succeeded to obtain many useful monoclonal antibodies specific ABO blood group and have established a methods for selective production of monoclonal antibodies to sugar moieties and peptide moieties of glycoproteins.
