Research Abstract

Natural killer (NK) cells possess two different kinds of receptors for the killing of target cells. One receptor transmits killing signals by binding to carbohydrate molecule on the target cells. Such second receptor as Ly-49A recognizes class II molecule of major hiotocompatibility complex on the target, and sends negative signal to NK cells. We have found that the expression of inhibitory receptor Ly-49A carrying ITIM sequence in the cytoplasm is under the regulation of DNA binding protein. Analysis of this DNA binding protein employing South Western technique revealed that the protein is identical to ATFII. We next examined whether this protein is expressed in rat kidney tissue or cultured rat mesangial cells. Interestingly, mRNA of ATFIL can be detected in both test samples. Considering that the expression of Ly-49A is lacked in kidney tissues, unknown sets of DNA binding protein other than ATFII is indispensable for the expression of Ly-49A in glomeruli. There is an increasing consensus that inhibitory receptors having ITIM sequence is regulated by the same DNA binding protein. We therefore explored whether receptor(s) carrying ITIM is expressed in rat kidney. For this purpose, we cloned mRNA from normal rat kidney that exhibits ITIM sequence. The product of the mRNA turned out to be Ly-49C, one of the family of Ly-49A and capable of transmit negative signal. These results indicate that ATFII in kidney functions as the key protein for the expression of Ly-49C. The behavior of the kidney cells may be maintained by the negative signals that are transmitted by Ly-49C.
