Research Abstract |
The utility of nested reverse transcription polymerase chain reaction (RT-PCR) using prostate specific antigen (PSA) and prostate specific membrane antigen (PSM) primers for prostate cancer staging was examined. 1. The nested RT-PCR for the PSA and PSM primers was able to detect one LNCaP cell per 10^6 leukemia (K562) cells. None of control subjects were found positive by the nested RT-PCR.2. The nested RT-PCR using the PSM primers appears to predict the prostate cancer stage more accurately than do nested RT-PCR using PSA primers and the conventional clinical staging modalities. 3. The nested RT-PCR for PSM assay was the best preoperative predictor of biochemical recurrence among the parameters examined. 4. Nested RT-PCR for PSM assay may provide a more accurate estimation of the true incidence of micrometastatic prostate cancer cells in lymph node than pathological examination and was a better predictor of biochemical recurrence than nested RT-PCR for PSA and pathological examination.
