Research Abstract |
To understand the function of cell proliferation-associated antigen, Gp125, which is a heterodimer of a 85-kDa heavy chain (CD98 : HG) and a 40-kDa light chain (LC), the following investigations were planned ; 1) Production of monoclonal antibodies against CD98, CD98-associated molecules and LC, 2)Molecular cloning of cDNAs for CD98-associated molecules and LC,3)Establishment and analysis of CD98-transfected cell clones, and 4)Establishment of CD98 transgenic mouse strains. 1) We produced monoclonal antibodies recognizing several epitopes on CD98 and light chain. Furthermore, we identified cytoskeletal toropomyosin as aCD98-associated molecule by the analysis with monoclonal antibodies. 2) We cloned cDNAs for LC and a truncated glutamate transporter (EAAC-1), which has partial amino-acid homology with LC. 3) We established and analysed wild-type, truncated and mutatedCD98-transfected clones, and demonstrated the relationship between the overexpression of CD98 and malignant transformation. 4) We established several transgenic mouse strains, in which human or rat CD98gene was stably intrgrated to the mouse genome.
