| Research Abstract |
In this project, the target substances of staphylococcal exfoliative toxine(ET) have been searching form mouse skins. The serotype A ET(ETA) has an activity to cut off the fibronectine of human plasma. We isolated 83 k Da fragment of fibronectine which treated with ETA. Amino acides sequence of N-terminal part in the fragment were analyzed. From these amino acid sequence, we made an oligoncleotide primer-5'-GCI CCI GCI GTI ACI GTI AGI TAI-3' (ETA5). The m RNA was isolated from skin of new born mice and cDNA was synthesized by the reverse transcripatase method. The cDNA was connected with Sau3AI casset and PCR was performed using primer of ETAS and casset primer CI. The 280bp band was obtained and their sequence was 3'-nontranslrated area of β-catenin. The new primers were made from genoms of β-catenin and plakoglobin. Using the new primers, c DNAs from new born mice and 10 days old mice were amplified by a PCR method. The amplified bands were shown in c DNAs from new born mice, and these were not shown in c DNA from 10 days old mice. The susceptibility of mouse shins against ET was depend on age, within 4 days old mice were high susceptibility and in 10 days old mice were poor susceptibility to ET. We are continue to research these relationship between susceptibility of mice skins to ET and gene expression of β-catenin and/or plakoglobin.
