Research Abstract |
To search for possible cancer chemopreventive agents, many K.ampo-prescriptions were assayed by primary screening indicated by inhibitory effect on Epstein-Barr virus early antigen (EBV-EA) activation in vitro. Of these prescriptions, Shouseiryu-to exhibited the significant inhibitory effects on EBV-EA activation, and exhibited strong anti-tumor-promoting activities on mouse skin and pulmonary two-stage carcinogenesis tests using TPA and glycerol, respectively, as promoters. Futher, this prescription showed the strong anti-tumor-initiating activity on mouse skin carcinogenesis induced by the NO donor. Searching for the active constituents of Shouseiryu-to, each component (eight crude drugs) of this prescription was assayed using EBV-EA activation test. Two lignans, asarinin and xanthoxyilol, were isolated from the MeOH extract of root of Asiasarum plant as the active constituents, and these lignans showed significant inhibitory effects on the two-stage carcino- genesis test in vivo. On the other hand, although these lignans were not isolated from the water extract of this crude drug alone but isolated from the water decoction of Shouseiryu-to prescription. Therefore, these lipophilic lignans could not extracted with water from single crude drug, but extracted from Kampo-prescription consisited with plural components. From these results, it was concluded that Shouseiryu-to might be valuable for anticarcinogenesis agents (chemopreventive agents) and one of the active compounds was the lignan. As a continuation of these studies, other crude drugs and folk medicines were investigated, and the anti-carcinogenic activities of several compounds were also reported.
