Research Abstract

個体発生の過程において、中枢神経系を構成するニューロン群が神経回路網を形成していくプロセスは、神経系の機能的構築を理解する上で、最も基本的な問題点の一つである。脊髄は、中枢神経系の中では比較的単純な細胞構築からなるが、機能的には複数の入力・出力が複雑に絡み合い、統合されている極めて高度な機能構造体である。われわれは、膜電位感受性色素とニューロン電位活動の光学的多チャネル測定法を発生初期胚脊髄に適用し、個体発生に伴う脊髄内神経回路網の機能形成過程について解析を行った。
孵卵8日の鶏胚脊髄スライス標本において、脊髄神経の刺激によって誘発される光学的シグナルは、刺激側のventral hornおよびdorsal hornの二つの領域から検出された。この二つの応答領域で、シグナルはそれぞれ速いスパイク状の成分(fast-signal)とそれに続く時間経過の遅い成分(slow-signal)からなり、このうちslow-signalは、外液Ca^<2+>依存性や疲労現象などの特性からシナプス後電位であることが明らかとなった。一方fast-signalは、外液Ca^<2+>除去によって影響されず、テトロドトキシンによって消失することから、活動電位に相当するシグナルであることがわかった。脊髄の細胞構築との対応から、ventralのfast-signalは逆行性刺激によって運動神経細胞に生じた活動電位であり、slow-signalは感覚神経の順行性刺激によって引き起こされたものであることが示唆された。dorsal hornとventral hornではslow-signalの波形や特性に違いが見られ、ventralの方が立ち上がりや時間経過が遅く、またシナプス疲労が速い、coolingからの回復が遅いなどの特性を示した。われわれは、光学的シグナルの時間的・空間的マッピングを行い、sensory neuron→inter-neuron→motoneuronにいたる脊髄神経回路網の活動パターンを、画像イメージとして捉えることに成功した。
