Research Abstract

本年度は以下の2つの実験を行った.実験には雄ウイスターラット(300-350g)を用い,侵害受容閾値の測定にはプランターテストを用いた.
[1] インターロイキン-1β(IL-1β)を視床下部視索前野(POA)に投与したときに生じる痛覚過敏におけるNOの関与の検討.実験1週間前に麻酔下でガイドカニューレとスタイレットをPOAの1.0mm上まで埋め込み,実験当日,薬物を目的部位に注入しpaw-withdrawal latencyの変化を観察した.IL-1β10pgをPOAに投与すると15-30分後にpaw-withdrawal latencyは短縮したが,IL-1β+N^G-monomethyl-L-arginin(10μg)を同時投与すると短縮効果が抑制された.POAにおいてIL-1βはNOを介して痛覚過敏を生じると考えられた.
[2] Lipopolysaccharide(LPS)全身投与によって生じる痛覚過敏/鎮痛作用におけるプロスタグタンジン(PG)の関与の検討.LPS1-100μg/kgを静脈内投与したところ,投与45-60分後にpaw-withdrawal latencyが短縮した.一方,120分の観察時間内ではpaw-withdrawal latencyの延長はみられなかった.シクロオキシゲナーゼ-2阻害剤であるNS-3981.0ng/0.3μlを両側POA内に局所投与したところpaw-withdrawal latencyの短縮が抑制されたが,腹内側核,室傍核内への投与では抑制効果がみられなかった.細菌感染症にかかった時に生じる痛覚過敏にPOAでのPG産生が関与すると考えられる.
