発癌におけるTGF-βシグナル伝達系関連遺伝子の役割

Research Abstract

近年、TGF-βシグナル伝達系の異常と発癌との関係が注目されており、その関連遺伝子であるTGF-βII型レセプター(RII)やSmad(Smad2とSmad4)遺伝子の異常が遺伝性非腺腫症性大腸癌(HNPCC)や一部の散発性癌で検出されている。本研究では、大腸癌の発癌機構を明らかにするため、HNPCCおよび散発性大腸癌でTGF-βシグナル伝達系関連遺伝子を調べた。
1) HNPCCにおけるTGF-βRIIの解析
1家系のHNPCCでTGF-βRIIの生殖細胞変異を検出し、この遺伝子がHNPCCの原因遺伝子の一つであることを明らかにした。
2) Smad3遺伝子の解析
Smad3はSmad2およびSmad4と同様に、TGF-βシグナル伝達に強く関連しているが、その異常と発癌との関係は不明である。我々は、Smad3遺伝子のゲノム構造を解析し、9個のエクソンからなることを明らかにした。散発性大腸癌とHNPCC腫瘍について、Smad3遺伝子を解析したが、異常は検出されなかった。従って、大腸癌の発癌におけるSmad3の関与は低いと推測した。
3) 大腸癌の発生とTGF-βシグナル伝達系関連遺伝子との関係
大腸癌発生は、腺腫から癌が発生する腺腫-癌説と正常粘膜上皮から直接癌が発生するdenovo説の2つに大別される。両者の代表的な例として、腺腫-癌型には腺腫内癌や隆起(ポリープ)型早期癌、de novo型には陥凹型早期癌が考えられている。そこで散発性の腺腫内癌、隆起型および陥凹型早期大腸癌についてTGF-βRII遺伝子を解析した。TGF-βRII遺伝子の異常はそれら3種類の癌ではほとんど見つからなかった。現在、それらの癌でSmad2およびSmad4の異常の有無を検討している。
