Research Abstract

RETプロトオンコジーン(以下RET)はGDNF(グリア細胞株由来神経栄養因子)、NTN等のGDNFファミリーをリガンドとする受容体型チロシンキナーゼをコードし、甲状腺髄様癌、副腎褐色細胞腫を遺伝的に発症する多発性内分秘腫瘍症(MEN)2A型、2B型、家族性甲状腺髄様癌及び大腸腸管神経節が欠損したために生じるヒルシュスプルング病(以下HSCR病)の原因遺伝子であることが明らかになっている。本研究ではHSCR病型変異を有するRET蛋白の不活化する機序を生化学的に解析した。
RETにHSCR病に認められたn種類の変異(E762Q,S765P,S767,F893L,R897Q,K907E,E921K,R972G,P973L,M980T,P1039Q,L1061P)を導入し、神経系細胞SK-N-MCに発現を試みた。1漏類のうち9種類のHSCR病型変異に(S765P,S767P,F893L,R897Q,K907E,E921K,P973L,P1039Q,L1061P)ついては、Ret蛋白の良好な発現を得ることができず、コントロール(c-Rct)の十分の一程度であった。10種類のHSCR病型変異を有するRct蛋白は蛋白レベルで不安定である可能性が示唆された。3種類(E762Q,R972G,M980T)の変異を有するRet蛋白は良好な発現を得ることができ、それらの発現しているSK-N-MC細胞とc-Ret蛋白が発現しているSK-N-MC細胞の間でシグナル伝達機構のどこに違いがあるのかを検討している。
