Research Abstract

細胞内寄生性細菌感染に対する防御は、ヘルパー1型T(Thl)細胞を活性化することが重要である。インターロイキン(IL-15)はIL-2と異なり、上皮細胞、マクロファージなど、広い組織において産生されるT細胞増殖因子であることから、感染局所におけるT細胞の活性化に深く関与している。本研究では、IL-15の感染防御機構における役割を明らかにするため、IL-15トランスジェニック(Tg)マウスを作製し、感染抵抗性に及ぼす影響について調べた。
RT-PCRでクローニングしたマウス(m)IL-15cDNAをトランスジーン用ベクターpHSE3'に組み込み、mIL-15Tgマウスを作製した。Tgマウスおよびコントロールマウスに、細胞内寄生性菌サルモネラを感染させ、経時的に臓器内菌数を測定した。また、腹腔侵出細胞をフローサイトメーターによって解析した。
Tgマウスにおいて、感染6日目に顕著な菌数の減少が認められた。また、感染3日目にはNKおよびNKT細胞が、6日目にはCD8陽性T細胞が著しく増加した。このCD8陽性T細胞の増加に伴って、血清中のインターフェロン(IFN)-γの上昇が認められた。さらに、TgマウスのCD4陽性T細胞のサルモネラ抗原特異的IFN-γ産生量を調べたところ、その活性が著しく高まっていた。
これらのことから、IL-15Tgマウスにおいて、Thl細胞への分化に関与するCD8陽性T細胞が増加し、感染抵抗性が増強していることが明らかとなった。
