Research Abstract

本研究では、NK細胞に異常をきたすマウスC57BL/6 nkd/nkd(以下nkdマウス)由来の神細胞を免疫原として、nkdマウス由来のNK細胞の細胞傷害活性を修飾するモノクローナル抗体Yuri 3を得、その性状解析を行った。Yuri 3抗体が認識する抗原の発現分布をFACSを用いて解析した結果、Yuri 3抗原は牌臓、骨髄、胸腺において全てのNKおよびNKT細胞に限局して発現することを見いだした。poly I:Cを腹腔内に注射したnkdマウスより調製した活性化NK細胞をエフェクターとして、Yuri 3抗体が細胞傷害活性に与える影響を検討した結果、Yuri 3抗体存在下ではsyngeneictargetに対する細胞傷害活性を誘導することを見いだした。このことはYuri 3抗体が認識する抗原が、NK細胞の標的認識に関与する可能性を強く示唆する。NK細胞の分化過程において、Yuri 33抗体は、胎児肝細胞内においてLy-49C/I陰性細胞画分に発現し、Yuri 3陽性細胞の割合の増加に伴ってLy-49C/I陽性細胞の割合は減少した。Yuri 3抗体が認識する分子が、神細胞分化の早期から発現すること、胎児肝細胞においてはLy-49C/Iと相補的な発現パターンを示すことから、この分子がNK細胞の分化過程で機能する可能性を検討している。
