Research Abstract

放射線照射とマイルドハイパーサーミア,およびメチルキサシチンの併用における照射効果の増感と細胞周期制御の関係についてヒト肺癌株を用いまず実験を行った.低線量率照射を41度の温度で行うと,照射効果の増感が認められた,照射のみではG2ブロクが著明であったが,照射と41度を併用すると,完全に増殖は抑制され,細胞はG1期に集積し放射線抵抗性のS期細胞が減少した.また照射併用時は熱耐性が完全に抑制された.同様に低線量率照射を1mMのカフェインを添加して行うと,照射効果の増感か認められた.カフェインの併用により,細胞はG1期に集積したが,このとき細胞数はむしろ増加し,照射によりG2期に集まった細胞の細胞周期を進行させ細胞死へ導いたものと考えられた.照射,41度,カフェインを同時に行うと細胞死はさらに増強し,細胞周期の変化は強いG1ブロックであった.ウエスタンブロットによるHSP72/73の解析では,三者の併用は治療時間内のHSP72/73の産生を抑制する結果が得られた.
また近年開発された抗腫瘍剤であるタキソールは,微小管重合を阻害し細胞をG2期に集積させる.これとメチルキサシチンを同時に作用させると,濃度依存性にタキソールの効果を阻害することが明らかとなった.メチルキサンチンによりG2期への細胞の供給が減少したためと考えられるが,詳細な細胞周期の解析やアポトーシスの関与などはまだ研究の途中である.
