Research Abstract

【1】 アポリポタンパク質A-l(アポ蛋白A-l)によるコレステロール引き抜き作用を阻害するモノクローナル抗体の分離
アポ蛋白A-lによるマウスマクロファージ由来泡沫細胞からのコレステロール引き抜き作用を阻害するモノクローン抗体の分離を試み、目的の阻害活性を有する3株のハイブリドーマを分離した。これらの培養上清からプロテインAカラムを用いて抗体を精製した後、抗原のマウス腹腔マクロファージの細胞膜画分に対してイムノブロットを行ったところ、バンドが認められた。現在、これらの抗体によって認識されるタンパク質の同定を急いでいる。
【2】 アポ蛋白A-lによるコレステロール引き抜きとコレステロールエステルサイクル
マクロファージ泡沫細胞に蓄積したコレステロールエステル(CE)は、中性CE加水分解酵素(NCEH)による脱エステル化と、acyl-coenzymeA:cholesterol acyltransferase(ACAT)による再エステル化の動的平衡(CEサイクル)にあり、この回転速度の大小が、HDLによるコレステロール引き抜き作用の一つの律速因子となっている。細胞膜アポ蛋白A-l受容体を介して引き抜かれる遊離コレステロールプールと、ACATの基質となる遊離コレステロールプールには共通部分が存在するため、本年度は特にACATにも注目して研究を進め、以下の知見を得た。ラットACATのcDNAをクローニングし、抗体を作成して組織分布を解析し、更にラット腹腔マクロファージでの活性調節機構を解析したところ、修飾LDLによって、mRNAの増加を伴わずにACAT蛋白量が増加し、酵素活性が上昇することが示唆された。現在、このメカニズムの解析、並びにHDLあるいはアポ蛋白A-lの効果を検討中である。
