| Research Abstract |
【目的】IgA腎症で有意に高い事が観察されているIgAlの自己凝集能およびmatrix蛋白への粘着能においてIgAl糖鎖の果たす役割をin vitroで検討すること。 【方法】(1)酵素的にNeuNAc、Gal、GalNAcを除去されたIgAlを作成し(asialo、agalacto、naked IgAl)、その分子量を電気泳動によりnative IgAlと比較した。(2)asialo、agalcto、naked IgAlの細胞外matrix蛋白に対する結合性をELISAによりnative IgAlと比較した。【結果】(1)asialo、agalacto、nakedIgAlはnative IgAlにはみられない分子量600kD以上におよぶ凝集物として存在した。(2)asialo、agalacto.naked IgAlはいづれもnative IgAlに比較して有意に強くtype IVcollagen、fibronectin、lamininに結合した。特にagalacto IgAlの結合能が最も強かった。【結論】糖鎖の除去はIgAlの自己凝集を惹起し、matrixへの粘着能を増加させた。IgA腎症におけるIgAl糖鎖異常の糸球体IgA蓄積機序への関与が示唆された。
