Research Abstract

PCNAは,真核細胞のDNA複製に関与するDNAポリメラーゼδ(polδ)のprocessive DNA合成に必須の補助因子であり,複製因子C(RFC)のDNA依存性ATPase活性を促進することが知られている.また,紫外線照射により損傷を受けたプラスミドを鋳型とした無細胞ヌクレオチド除去修復系による実験から,ヌクレオチド除去修復因子の1つとしても示唆されている.さらに,サイクリン依存性キナーゼの阻害因子であるp21やGADD45,DNA複製および修復の両方に関与するFEN-1等に結合することが報告される等,PCNAはDNA上で起こる各種反応の制御機構に重要な役割を果たしていると考えられている.本研究の代表者は,PCNAが関与するDNA修復の分子機構の解明を目的として,組換え型ヒトPCNA変換体を用いて,特にFEN-1との相互作用の解析を行っている.以下に,今年度得られた新たな知見等の成果を示す.
PCNAの21番目のアスパラギン酸をアラニンに変換した変異体ではRF-cの.ATPase活性促進およびPolδのDNA鎖伸長反応促進は野生型PCNAと同様であったがFEN-1のエンドヌクレアーゼ活性促進は野生型PCNAに対し20〜40%まで低下していた.そこで,21番目のアスパラギン酸をグルタミン酸(D21E)またはアスパラギン(D21N)に変換した変異体を作製し,FEN-1のエンドヌクレアーゼ活性促進能を調べたところ,D21Eでは野生型と同等の活性促進能を示したが,D21Nでは野生型の10〜30%しか活性促進能がみられなかった.以上のことから,FEN-1のエンドヌクレアー活性促進にはPCNAの21番目のアスパラギン酸の負電荷が重要と考えられた.
