Research Abstract

現在、心筋梗塞や脳血栓などの血栓症による疾病が急増し、ストレプトキナーゼなどの治療薬が用いられているが、抗体形成などの問題点があり、血栓症を防ぐ安全な抗トロンビン活性物質の探索が望まれている。本研究では、血栓症の予防として、その原因である血栓の形成を抑制する血液凝固抑制物質(抗トロンビン活性物質)および血栓を溶解する線溶酵素などの血液関連生理活性物質を生産する微生物を用いて、新しい食品素材の開発を行うと共に、食品への応用を試みた。したがって、本研究の主目的は、心筋梗塞や脳血栓などの血栓症に予防効果を示す機能性食品の開発である。本申請者はこれまでにクモノスカビの培養液中に抗トロンビン活性物質を見出し、フマル酸であることを明らかにし、フマル酸は、清酒、味噌、醤油、甘酒など発酵食品製造に用いられるコウジカビにおいても生産されることから、発酵食品の摂取は血栓症の予防につながる可能性を示唆した。本補助金により、まず、これまでに見出した全く新しいタイプの抗トロンビン活性物質を生産する野生種マスタケからの本活性物質の分離・精製を試みた。また、線溶酵素活性について種々の微生物からスクリーニングを行い、タモギタケや野生種担子菌W510などにその著しい酵素活性を見出した。さらに有効成分を有する野生種担子菌などについては、有用担子菌の育種を目的として、プロトプラスト化、パールチェーン形成、細胞膜の一過的破壊などの電気融合のための諸条件を検討すると共に、効率の良い人工栽培のための諸条件の検討を試み、抗トロンビン活性物質を生産し人工栽培が可能な食用担子菌の育種に成功した。さらに、心筋梗塞や脳血栓などの血栓症に予防効果を示す機能性食品を開発した。今後はさらに、これら育種した融合担子菌や機能性食品の諸性質を明らかにすると共に、一般的食品化への応用を試みる予定である。
