Research Abstract

1、 コルチコステロン投与ラットにおける脳神経細胞の神経機能陣審の検討
7週齢のWiatar系雄性ラットにナタネ油に溶解したコルチコステロン40mg/5ml/kgを投与した。10日間の連続投与後.潅流固定を行い、バラホルムアルデヒドで固定後、凍結切片を作成した。この切片に対して、シナプトフィジンやシンタキシの細胞骨格あるいはカルシムチャンネル形成タンパクの免疫染色を行い、光学顕微鏡で観察した.しかし、各タンパクの局在化や染色強度に変化は観察されなかった.
2、 ニューロトロフィンタンパクの高感度酵素免疫測定法の確立
培養神経細胞上清や細胞内のニューロトロフィン含量を検討するためには、各々のタンパクを裏感度に測定する必要がある.今までニューロトロフィンタンパクの裏感度の測定系は確立されていなかった.我々は、ニワトリ抗血清を用いることにより、非常に高感度の脳由来神経栄養因子およびニューロトロフィン-3の測定系の確立に成功した.
3、 培養ラット脳神経細胞におけるニューロトロフィンの発現に対するコルチコステロンの影響
胎生18日のラットから取り出した海馬神経細胞にコルチコステロンを24作用させた後、細胞中と上清中のニューロトロフィン含量を測定した.ニューロトロフィン-3の含量に関してはいずれもコルチコステロンによる有意な変化は観察されなかったが、細胞内の脳由来神経栄養因子の含量は減少していた.
