Research Abstract

本年度は昨年度に引き続きプルキンエ細胞の樹状突起形成期に特異的なcDNAサブトラクトライブラリーの構築とクローンの単離を目指した。また、この研究計画遂行中に非必須アミノ酸であるL-セリンがプルキンエ細胞や海馬錐体細胞の強力な生存促進因子であることを見い出したことから、中枢神経系でL-セリン輸送に関わると推定されるトランスポーター分子をRT-PCR法で単離することを試みた。その結果、これまでにセリン輸送能を持つことが知られているトランスポーターASCT1以外に、脳では発現していないと報告されていたsystem B-like中性アミノ酸トランスポーターの新しいサブタイプを培養小脳神経細胞BNAから初めて単離することに成功し、その構造解析を行なった。(論文投稿準備中)。このcDNAクローンをHEK細胞に導入して安定発現株を得ると、セリンなどの中性アミノ酸輸送活性がベクターのみの対照に比べ4-6倍増加していた。さらに、この分子のユニークな基質特異性としてグリシンの輸送活性を持つことも見い出した。また発達過程での発現パターンをNortern Blottingによって検討すると、出生直後に最も高い発現が観察された。以上の結果から、この新規トランスポーター分子が発達中の中枢神経系において中性アミノ酸輸送を担っているものと予想された。現在この新規トランスポーターとASCT1の脳内での発現分布をin situ hybridizationによって詳細に比較検討中である。また樹状突起形成期特異的クローンの単離と構造解析も平行して継続中である。
