渦鞭毛藻を用いた真核生物間の細胞内共生の実験的解析

Research Abstract

細胞内共生を見るために培養を行なった渦鞭毛藻Alexandorium tamarenseの培養が困難を極めた.したがって,渦鞭毛藻に関する解析は今回は行なうことができなかった.
細胞内共生の最初の過程は,異種の生物の相互認識に始まると考えられる.この過程で最も重要な機能を果たすのは,確証はないが他の細胞の例から類推して,糖鎖と考えるのが妥当である.そこで,細胞表面の糖鎖の特性を見るために,ラフィド藻のChattonella marinaを使って,その感受性を検討し,渦鞭毛藻への適用のための基礎データとした.
14時間明期,10時間暗期の条件で生育させた細胞に,糖鎖認識部位の異なる4種のレクチンを添加してその影響をヴィデオによる顕微鏡下での連続観察によって検討したところ,細胞の溶解,運動性の低下などが観察された.さらに詳細にみると,溶解する細胞の多くは分裂直後の細胞である事が判明した.レクチンにより細胞の形が変わる(楕円形から桿形へ)細胞は分裂後数時間を経たもの,まったく感受性が認められない細胞は栄養成長の状態にあることが判明した.またレクチンの種類から感受性に関与する糖鎖は,N-acetylglucosamineである可能性が高くなった.これらの結果は,細胞周期にしたがって細胞表面に存在する糖鎖の量が関連していることを示し,まったく予想をしなかった画期的な結果を得た.
こうした観察に基づいて,今後は細胞周期と細胞表面の糖鎖の質的,量的変動を解析し,微細藻類の細胞表面の認識機構をさらに追及する予定である.
