Research Abstract

昨年度、1)マウスにおいて、human proinsulinはhuman insulinに対し1/50程度のbioavailabilityを保持することを明らかにし、2)ヒト膵組織より作製したpreproinsulincDNAのconstructを挿入したretrovirus vectorを作製し、producer細胞を得た。
それに引き続き、今年度は、moloney白血病ウイルス由来のレトロウイルスベクターpGK,pRxにhuman insulinおよびコントロールのconstructを挿入して作成した、合計4種類のretrovirus vectorを、lipofection法を用いてpackaging細胞にtransfectし、G418によるselectionを行って、各々約20クローンのproducer胞株を樹立した。pGk vetorはNeO耐性遺伝子を持たないため、Neo遺伝子をもったplasmid vector(pRc/CMV)をco-transfectした。
producer細胞の一部のcloneについてプロインスリン/インスリン抗体を用いた免疫染色を施行した。pGk vectorをtransfectしたproducer細胞では、挿入した配列よりimmunoreactive proinsulinが産生されることが確認された。また、培養上清のインスリン濃度をEIAL法により測定し、immunoreactive proinsulinの産生を確認した。
