| Research Abstract |
本研究において逆転写酵素に会合したRibonuclease H活性を測定するための3つの方法を確立した。
(1)poly(rA)・poly(dT)の標識体を用いてRNAの分解を測定する方法
(2)オリゴヌクレオチド(dGTCATCTCCとrGGAGAUGAC)を基質として用いHPLCにより分解を測定する方法
(3)蛍光標識したオリゴヌクレオチドの基質分解にともなう蛍光強度の増強をマイクロプレートリーダーを用いて測定する方法(fuluorescence energy transfen法)
これら測定法は一長一短があり,多数の天然物エキスのRNaseH阻害作用を調べるためには(1),(3)の方法がすぐれていることを実証した。
Juglans mand shuricaからRNaseH活性を強く阻害する成分の探索を行った。
また,種々の天然に存在するキノン類のRNaseH阻害作用を検討し,ナフトキノン類に強い阻害作用を認めた。
