Research Abstract

小型化抗体遺伝子を植物へ導入することにより、抗体と2次代謝産物とのコンプレックスを生成し、系外へ除去されることにより2次代謝産物の生合成経路のキャパシティーが増大し、より多量の2次代謝産物を生産蓄積するというアイディアに基ずくものである。
Agrobacteriumに組み換えるために、まず大腸菌でのscFVの発現系を構築した。抗forskolinMAb産生ハイブリドーマからmRNAを抽出し、逆転写によりcKNAを合成したVHとVLの可変部分に相補的なプライマーを用いてPCRを行いVH,VLを増幅した。VH,VLをペプチドリンカーにより連結しscFV遺伝子を構築する。このものをpET-28aベクターへ組み換えた。本ベクターを大腸菌BL21に形質転換し大量発現系を構築し、IPTGの添加によりscFVを発現させた。scFVはin clusion bodyとして発現するため尿素で可溶化し希釈法により巻き戻した後His-tagを用いたアフィニティーカラムにより精製を行うことによりscFVタンパクを得ることが出来た。本法によれば培地100nl当たり約6mgのscFVが得られる。現在までに抗forskolinMAb scFVとforskolinとのモデリングを行っている。現在scFV遺伝子を、Agrobacteriumに組み換え中で、今後forskolin含有Coleus horskohliiに組み換え植物体で発現させることによりforskolinの生合成をコントロールし、forskolin高含有株を作出すべく実験を進めている。
