Research Abstract

【研究の目的】
本研究は、ヒトモデル系として期待されるブタにおいて、下痢症に関わる免疫パラメータを指標として、イムノバイオティクスの選抜から、その実証研究を行うことを目的としている。本年度は、昨年度までの成果を基礎として、イムノバイオティック機構の解析を進め、選抜イムノバイオティクスの有用性について検討した。
【研究成果】
1.イムノバイオティクスのPIE細胞における抗炎症シグナル伝達機構:選抜イムノバイオティクスによる細胞内シグナル分子の活性調節について解析したところ、MAPKのリン酸化とIkB分解の抑制が認められ、それらに関連するネガティブ調節因子の発現誘導が見られた。2.腸管パイエル板モデル培養系による抗炎症機能解析:PIE細胞と腸管パイエル板由来免疫担当細胞との共培養系において、抗炎症性発現における細胞間のサイトカインネットワークの特徴が把握できた。これらの結果から、選抜イムノバイオティクスの腸管抗炎症に関わる調節機構の一端が解明できた。今後、インビボ実証試験を続けることにより、選抜イムノバイオティクスの有用性が検証できる。
