Research Abstract

前年度までに、PHD2のsiRNAによるノックダウンによって、マウス下肢虚血モデルの治療が可能になることが確認されたために、本年度は、HIF-1αを制御する他の因子に関して検討を行った。
具体的には、前年度までの研究において転写因子Ying Yang 1(YY1)がHIF-1αを正に制御していることが確認されたために、本年度は、siRNAによるYY1のノックダウンに基づくがん治療の可能性に関して検討を行った。YY1は、がん細胞で過剰発現しており、p53の発現量を負に制御することが報告されており、siRNAによるYY1のノックダウンは、がんの分子治療の有効なアプローチとなりうる。研究の結果、低酸素でYY1を抑制することによりHIF-1□タンパク質の減少、HIF-1転写活性の低下及びHIF-1標的因子発現の減少がもたらされることが確認された。
これらの結果は、低酸素状態でYY1がHIF-1□を制御することを示唆している。これに関して、免疫沈降によって、YY1とHIF-1□の直接的な相互作用も確認されている。
一方、YY1に対するsiRNAを安定発現させたがん細胞を用いて肺転移モデルを作成したところ、YY1のノックダウンによってがんのコロニーが減少することが確認された。
また、YY1のノックダウンによるHIF-1の抑制はp53非依存的であることも確認された。これらの結果より、YY1はp53に非依存的に悪性腫瘍に対して治療効果をもたらすことのできる有望な治療ターゲットになるものと考察された。
