Research Abstract

光合成反応中心複合体の環境適応過程の解明を目的とし、Prochlorococcus marinus(P.marinus)の先祖がモノビニルクロロフィル(MV-Chl)のかわりに新たな光合成色素としてジビニルクロロフィル(DV-Chl)を獲得した際にphotosystem II(PSII)反応中心(RC)にどのような変化が必要とされたのかを明らかにした。前年度にはPSII-RC由来の遅延蛍光スペクトルを解析することで、RC近傍での変化を明らかにできることを初めて報告した。今年度は野生型でW-Chlを持つSynechocystisに、1)DV-Chlを導入した変異体、2)さらにPSII-RCの二ヶ所のアミノ酸をP.marinusタイプに置換した変異体、の遅延蛍光スペクトルの解析を行った。色素がDV-Chlに変化することで、PSII-RCに接続するCP47内部でカロテノイドに近接しているレッドクロロフィル(red Chl)へのエネルギー移動経路が一部阻害され、RCに励起エネルギーが集中していた。これは弱光環境においては有利な形質であり、P.marinusの先祖が貧栄養の弱光環境に適応するためにDV-Chlを活用した可能性を示唆する。一方でこの変異体は強光下では死滅した。ここでPSII-RCのアミノ酸をP.marinusタイプに置換することで強光耐性が回復した。この際、PSII二量体内部でRCを迂回する形でCP47のred Chlへの速いエネルギー移動経路が新たに形成されており、この経路は過剰なエネルギーを効率的に熱として散逸させるのに有用だと思われる。P.marinusの先祖は、貧栄養の弱光環境に適応するためにDV-Chlを獲得した後に、一時的な強光にも対応するためにPSII-RCの二ヶ所のアミノ酸を最適化させることで部分的な強光耐性能力をも獲得したと考えられる。
