水銀によるヘム制御時計遺伝子転写の協奏の障害と一酸化窒素及びチオールによる修復

Research Abstract

哺乳動物の前脳に存在するヘム結合時計遺伝子転写制御因子と推定されマウスPer1,マウスPer2,及びマウスPer3のヘム結合様式、チオールの関わり、水銀感受性、及び一酸化窒素感受性などを調べ、水銀により引き起される疾病の中で、時計遺伝子に関わる疾病の修復の可能性を調べることを目的にする。[1]マウスPer1,及びマウスPer2のN末端の存在するPASドメインのチオレートがヘム結合部位であることが示唆された。[2]マウスPer1,及びマウスPer2へのヘム結合は水銀により解消され、さらに過剰のチオレートを添加することにより修復された。[3]部位特異的変異法により、マウスPer2のシステインは同定が可能であったが、マウスPer1のシステインの位置を決定することは出来なかった。[4]ヘムはマウスNPAS2とマウスPer1の間を移動することが示唆された。[5]ヘムはマウスPer2のC末端側にある2つのCPモチーフの1つのシステインに結合することが示唆された。この場所は人由来Per2のCPもチーフとは異なる。[6]ヘムはマウスPer2の2つ目のPASドメインに結合するが、その結合部位はシステインではなくて、イミダゾールであることが示唆された。[7]マウスPer3はヘム結合性蛍白質であることが示唆されたが、ヘム結合の定量化は困難であった。[8]マウスへメチル水銀を添加することにより異常な行動が惹起された。しかし、チオレート試薬を血液脳関門促進試薬と同時投与した場合に、異常行動は修復された。この結果は、脳に存在する蛋白質のチオレート基に水銀が結合して異常行動を引き起すが、過剰のチオレート試薬により水銀が離脱し、異常行動が修復されたのではないかと推定された。
