Research Abstract

本年度は、オオバコモザイクウイルス(PlAMV)の複製精度に大きな影響を及ぼすと考えられる、PlAMVが引き起こす全身壊死(systemic necrosis)について解析を行った。PlAMVにコードされるタンパク質を、全身壊死を引き起こす宿主であるタバコ属のモデル植物Nicotiana benthamianaにおいてアグロインフィルトレーション法により一過的に発現させたところ、複製酵素が壊死を誘導することがわかった。細胞間移行タンパク質など他のタンパク質は壊死に影響を及ぼさなかった。また、誘導発現系を用いて複製酵素と壊死の関係についてさらに解析した結果、複製酵素の蓄積量が多いほど、壊死が強く誘導されることが示された。さらに、複製酵素における壊死誘導活性を持つ領域を絞り込んだ結果、複製酵素の中央部に位置するヘリカーゼドメインが壊死誘導活性を持つことを明らかにした。さらに、ヘリカーゼドメインによる壊死誘導活性は、無病徴の分離株にも見出されることは、上記の結果とあわせて、無病徴の分離株も蓄積量によって壊死を誘導する可能性があることを示唆しており、ウイルスの複製精度の低さに起因するゲノムの多様性と、病原性の進化について考察する意味で非常に意義深い知見である。
さらに、PlAMVとは異なるRNAウイルスの感染植物内でのゲノムの多様性を比較するため、さまざまなRNAウイルスのゲノム塩基配列の解析を行った。特に、PlAMVと同様ひも状のウイルス粒子で単一の一本鎖RNAゲノムを持ち、ウメに大きな被害を及ぼすウメ輪紋ウイルス(plum pox virus:PPV)の日本における遺伝的多様性を示した研究成果は、ウイルスの伝播・発病と遺伝的多様性の関係を示す知見であり、植物RNAウイルスの複製精度の低さに起因する多様性がエマージングウイルスの与える被害に影響する可能性を示唆している意味で興味深い。
