Research Abstract

当該年度は、2つの大きな成果をあげ、本研究の最終目標を達成することができた。
1つ目は、急性肝炎患者由来AH1株(遺伝子型1b)の全長HCVRNAの上流にレポーター遺伝子であるレニラルシフェラーゼを組み込むことにより、簡便に薬剤アッセイができるシステム(AH1R)の構築に成功した。このアッセイシステムを国際誌(Virus Genes)に投稿して、受理された(Mori K et al.,Virus Genes,2012)。AH1RとOR6(遺伝子型1bHCV-0株由来のアッセイシステム)を用いて、IFN-α、IFN-γおよびサイクロスポリンAの薬剤感受性を比較したところ、AH1株由来の全長HCVRNA複製システム(AH1細胞)とHCV-0株由来の全長HCVRNA複製システム(0細胞)を比較した以前の結果と同じ結果を示したことから、これらの薬剤に対する感受性の違いは、ウイルス側の要因で決まっていることが示唆された。
2つ目の成果は、本研究の最終目標であった「新しいLi23細胞を用いた、HCV生活環の再現システムの構築」を達成したことである。治癒細胞であるORL8cを用いて、感染性粒子産生系JFH-1(遺伝子型2a)の感染が認められ、さらに、ORL8cから産生されたJFH-1粒子がさらにORL8cに感染性を示すことを見出した。この結果により、既存のHuH-7細胞と異なる新しいLi23細胞におけるHCV生活環が完成したことになる。HCV生活環の再現システムを2種類の細胞で構築したことは、「HCVの感染経路の研究」および「HCVに関連する宿主因子の研究」などに広く応用でき、HCV研究においてとても意義のある成果である。
