Research Abstract

ゲノム・プロテオーム解析によって得られた大量の遺伝子機能情報や代謝経路情報に基づき細胞の全体または一部の代謝活動をコンピュータ上で再構築するための汎用のシミュレーションソフトウエア「E-CELL」を開発している。
E-CELLはSubstance-Reactorモデルに基づいており、酵素反応や膜輸送などの化学反応をReaction Ruleとして個々に定義するとそれらを疑似並列に実行し全体の振る舞いをシミュレートする。ユーザは物質濃度の増減や特定の反応の速度などを観察することができるだけでなく、グラフィックインターフェースから介入して任意の物質濃度を人為的に増減させる事もできる。
昨年までに我々はE-CELLシステムを用いて127個の遺伝子からなる仮想の「自活細胞モデル」を完成させたが、現在これに細胞増殖に必要な遺伝子を加えることによって「自活増殖細胞モデル」の構築を目指している。また、転写制御やDNA複製などの代謝を拡張するためにDNAや転写翻訳機構のモデリングに改良を加えている。一方、これらの仮想細胞モデルとは別に、E-CELLを用いてヒト赤血球細胞のシミュレーションやバクテリアchemotaxisのシグナル伝達のシミュレーションの完成も目指しており、特に赤血球細胞ついては、代謝全体のモデルが完成し、現在デバッグや改良を行なっているところである。
