Research Abstract

網膜芽細胞腫蛋白質(pRb)は腫瘍抑制遺伝子産物として細胞周期の制御に必須な働きをしている。我々の以前の研究によりpRbはその非リン酸化型が73kD heat shock cognate(HSC73)と複合体を形成することが示されているが、今回この意義につき解析を加えた。その結果、凝集した非リン酸化pRbは20Sプロテオゾームによりただちに分解されるが、リン酸化pRbはこれに抵抗性であること、HSC73はHSP40と共用してpRbの凝集および20Sプロテオゾームからの分解を阻止していることなどが明らかになった。これらの事実はHSC73が非リン酸化pRbの選択的な分子シャペロンとして重要であり、ひいてはpRbの機能発現にHSC73が必須な働きを行っていることを意味している。
