Research Abstract

本年度の研究成果は以下の通りである。
1.細胞質型PLA2(cPLA2)欠損マウスの表現型の解析に関する研究
cPLA2欠損マウスの表現型の解析を、成人呼吸窮迫症候群および大腸ポリポーシスについて行ってきたが、現在、脳虚血後再灌流傷害、ブレオマイシン気道投与による急性肺炎、コラーゲン惹起関節炎、アレルギー性脊髄炎などの疾患モデルの解析が進んでおり、いずれも本マウスにおいて症状が軽減していることを確認している。
2.新規PLA2に関する研究
カイニン酸によって痙攣発作を誘発したマウスやラットにおいて、海馬歯状回に時期特異的に発現するcPLA2遺伝子由来の新規蛋白質を発見し、そのプロモータ領域の解析を行っている。この新規蛋白質を発現する細胞が未分化の神経細胞であることを確認した。
3.生理活性脂質産生制御に関する研究
cPLA2と同時にカルシウム刺激に応じて核膜に移行してロイコトリエン産生に関与する5-リポキシゲナーゼの、細胞内局在と移行の分子機構に関する研究を行い、核移行シグナルと核外移行シグナルに依存した機構が存在することを明らかにした。
4.生理活性脂質受容体に関する研究
血小板活性化因子(PAF)受容体を介するPAFの分解を報告した。第2LTB4受容体のリガンドの性質について報告した。さらに、ペプチド性ロイコトリエン受容体のマウスにおける発現を解析し、種差や系統差による生理活性脂質の作用の差について考察を加えた。
