Research Abstract

ATP感受性K^+(K_<ATP>)チャネルはABC蛋白であるスルホニル尿素受容体(SUR)と内向き整流性K^+チャネルの複合体である。申請者らはこれまでに膵β細胞型K_<ATP>チャネルのKir6.2サブユニットのノックアウト(KO)マウスを用いて、黒質網様部のK_<ATP>チャネルが代謝ストレス時の全身けいれん発症の抑制に重要であることを明らかにしてきた。平成14年度には、1)麻酔下のKir6.2 KOマウスから、低酸素負荷時の呼吸活動と横隔神経活動を同時記録することにより、野生型(WT)マウスは正常呼吸型からあえぎ呼吸(gasping)型へと極めて短時間で移行するのに対して、KOマウスではgaspingではなく正常呼吸型のリズムを変化させて対応する傾向を見いだした。従って、膵β細胞型K_<ATP>チャネルはgasping形成に重要な働きを果たしている可能性が強い。
2)血管平滑筋型K_<ATP>チャネルのKir6.1サブユニットのKOマウスを千葉大学の清野進教授より供与を受け、使用可能になった。今後、脳局所における神経活動に伴う血流変化におけるK_<ATP>チャネルの役割についても検討を加える予定である。
3)申請者らが単離したABCA2のオリゴデンドロサイトにおける発現が、ラット脳の発達段階において、ミエリン形成とよく一致することを明らかにした。従って、ABCA2はミエリンの主構成成分である脂質の膜輸送に関連している可能性が強い。
4)ABCA2遺伝子のホモKOマウスを樹立した。
5)肺胞II型細胞に発現するABCA3の蛋白発現が、サーファクタントの合成と同様に、ラットの出生直前に急激に増大すること、グルココルチコイドにより発現が誘導されることを明らかにした。この結果は、ABCA3がサーファクタントの主たる構成成分である脂質の輸送に関していることを強く示唆している。
