Research Abstract

CFTRは欧米で非常に多い常染色体劣性遺伝疾患である嚢胞性線維症の原因となる膜蛋白である。この膜蛋白は、構造上はABCトランスポータのファミリーに属しながら、機能的には細胞内のcAMP増加によって活性化されるClチャネルの実体であることが明らかにされてきた。しかし、はじめの命名通り、他のチャネルに対するレギュレータとしての機能も併せ持つことが知られている。さらにパッチクランプ法によりCFTR強制発現細胞においてATP電流が観察できたという報告もあり、ATPの通過路はCFTR自体であるという仮説が米国の研究者から提唱された。更に最近、CFTRではなく容積感受性Clチャネルを通ってATPが放出されるという仮説や、放出されたATPが容積感受性Clチャネルの活性化の原因因子であるとの仮説も外国のグループから提出された。そこで本研究では、1)ATP放出はCFTR Clチャネルを介して行われるか?、2)ATP放出は容積感受性Clチャネルを介して行われるか?、3)細胞膨張時に放出されるATPは容積調節を実際に制御するか?、4)放出ATPは容積感受性Clチャネル活性化の原因因子か?、5)CFTRはATP放出を制御するか?、6)するとすればその分子メカニズムは?即ち、ATP放出路蛋白とCFTR蛋白の分子的相互作用機序は何か?、7)CFTRは容積感受性Clチャネルを制御するか?、8)するとすればその分子メカニズムは?、即ち、CFTR蛋白と容積感受性Clチャネル蛋白の分子的相互作用機序は何か?、9)結局CFTRは細胞容積調節を制御することができるのか?、そして10)このATP放出路の分子実体は一体何なのか?、という計10項目の設問に答える。これまでの4年間の研究によって、上記設問の内の7項目(設問1〜5,7,8)に解答を与えることができた。
今年度は残り3項目について実験的検討を行った。CFTRによるATP放出に対する制御の分子メカニズムの解明(設問6)は各種のCFTRミュータントを作製することによって行ったところ、NBD2のATP加水分解能をもったコンフォーメーションがATP放出路蛋白とCFTR蛋白の分子的相互作用に不可欠であることが判明した。CFTRの容積調節への制御的役割(設問9)は、CFTR発現細胞の方で、浸透圧性膨張後の容積調節能が高いことから明らかとなった。ATP放出路の分子実体(設問10)については、すでに発見したATP透過性マキシ・アニオンチャネルのCFTR依存性、容積感受性、薬理的感受性などの諸性質を調べ、これらが機能的にATP放出路に対応することを確定した。
