Research Abstract

リン酸イオンは、植物成長における三大栄養素の一つである。自然界で植物が利用可能なリン酸の土壌含有量は極めて小さいため(通常10μM以下)、多くの植物は常にリン酸飢餓状態にある。植物が生理機能を正常に遂行するためには、細胞質リン酸濃度を一定の値に保つ必要がある。この濃度維持機構をリン酸ホメオスタシスと呼び、細胞質のリン酸濃度を一定に保つために、液胞がリン酸のreservoirとして緩衝機能を持つことが知られている。本研究では、液胞がリン酸を蓄える機構および細胞質の必要に応じて液胞からリン酸を放出する機構がどの様に制御されているかを、特に液胞膜のリン酸輸送機構と細胞質のリン酸濃度認知機構に焦点を当てて明らかにすることを目的とする。
この4年間のニチニチソウ培養細胞の単離液胞を用いた研究で、液胞のリン酸取り込み活性が、ATP、PPiによる液胞膜のエネルギー化に依存し、膜電位依存のチャネル輸送体を通して行われることが明らかになった。すでにこの輸送系の分子実体の解析を開始し、輸送体に共有結合すると思われるリン酸取り込み阻害剤を見いだし、その阻害剤をマーカーにリン酸輸送体の同定を進めている。本年度は、ゲノム情報が明らかになっているシロイヌナズナ培養細胞からの液胞単離条件を決定し、シロイヌナズナでも同様にリン酸輸送体の分子解析が進められることを明らかにできたので、液胞膜のプロテオーム解析が可能になるように膜標品の確保につとめるとともに、かずさDNA研究所の佐塚博士、RITEの横田博士、嶋岡博士の協力を得て、膜タンパク質のプロテオーム解析を可能にするための条件検討を行った。現在、一次元SDS泳動標品から質量分析計を用いてタンパク質の同定をするための可能性を検討している。
さらに、車軸藻細胞を用いて、液胞膜・細胞膜のリン酸放出活性の実験を開始し、細胞膜からのリン酸放出に外部リン酸の存在が必要なことを見いだした。これについては、現在詳細を検討中である。
