Research Abstract

液胞膜タンパクのカタログ化
シロイヌナズナ緑色植物体から液胞膜濃縮画分を調製し、強膜結合タンパク質に対するプロテオーム解析を行った結果、96個の遺伝子が特定された。これらの分子の正確な細胞内局在と機能解析を行うために、約50分子に対する抗ペプチド抗体を作製した。自らの研究に使用するだけでなく、抗体を膜タンパク質研究者の実験ツールをして広く利用してもらうために、作製した抗体の検定を詳細に行っている。すなわち、抗体の特異性をELISA,生体膜試料との反応性、抗原ペプチドとの競合、抗体の使用濃度による反応性の変化などで検定している。一部の分子は既に抗体をオルガネラ膜分画に反応させ細胞内局在を調べた。その結果、液胞膜、細胞膜および小胞体膜に局在する分子が特定された。
液胞膜Ca^<2+>/H^+対向輸送体
植物液胞へのCa^<2+>、Cd^<2+>、Mg^<2+>、Mn^<2+>等の金属イオン集積機構を解明するために、イネ液胞膜Ca^<2+>/H^+対向輸送体の5種(OsCAX1a,1b,1c,2,3)をcDNAクローニングした。いずれも411〜453個のアミノ酸で構成され11個の膜貫通推定領域をもつ。機能検定をするため、酵母に個々の遺伝子を導入し発現させ、各種イオン耐性を検定した。その結果、Ca^<2+>のみを輸送する分子種、Mn^<2+>とCa^<2+>の両方を輸送する分子種があることが明らかになった。さらに、イオン選択性を司る機能ドメインについて、部位特異的変異導入法により、分子内繰り返し配列内のアスパラギン、アスパラギン酸、グルタミン酸、ヒスチジン残基が必須であることが明らかにされ、それらの膜内配置も検定した。
ダイコン水チャネルの水輸送機能の解析
ダイコンの液胞膜水チャネルの機能を検定するために、その遺伝子を酵母に導入して膜を調製し、ストップドフロー法で膜の水透過率を測定した。その結果、液胞膜型2種での水輸送活性を検出することができた。
