Research Abstract

昨年、分裂酵母の全ゲノム塩基配列が明らかにされた。そこで分裂酵母全ゲノムのデータから以下の遺伝子を同定して機能解析を行った。(1)分裂酵母の全ゲノムから出芽酵母の液胞タンパク質輸送に関与するVPS遺伝子と相同性の高い遺伝子を同定して、これらの遣伝子破壊株を50以上作成した。多くの破壊株で液胞タンパク質が細胞表層へとミスソートしていることを確認出来たが、全く表現型を示さない株も見いだされ、出芽酵母との相違点を明らかにすることが出来た。(2)分裂酵母の液胞局在ATPアーゼの欠損株を取得した。本破壊株は液胞の酸性化に欠損を示し、液胞タンパク質輸送やエンドサイトーシスなどの細胞内タンパク質輸送経路に欠損を示すことを明らかにした。(3)分裂酵母の小胞輸送に関与する16個の全SNAREタンパク質をコードする遺伝子を単離して、機能解析を行った。その結果、出芽酵母のどのSNAREとも高い相同性を示さないSPAC6F12.03C遺伝子破壊株が液胞タンパク質を細胞表層へとミスソートすることを明らかにした。(4)分裂酵母の液胞の融合に関与するタンパク質の同定を試みた。その結果、出芽酵母のVPS16とVPS33と相同性の高い分裂酵母遺伝子破壊株は液胞形態形成に欠損を示すが、VPS11やVPS18ホモログ遺伝子破壊株は液胞形態に欠損を示さないことがわかり、出芽酵母との相違点を明らかにすることが出来た。(5)分裂酵母のエルゴステロール合成に欠損を示す株を単離し、解析を行った。その結果エルゴステロール欠損株は液胞の融合が起こらないことがわかり、ステロールの液胞形態形成への関与が強く示唆された。
