Research Abstract

液胞は植物が行う生命活動における重要な機能を司ることが分かっているが、植物二次代謝産物、とくに花の主要色素であるアントシアニンの液胞への輸送と貯蔵、呈色に関する遺伝子レベルでの解析は進んでいない。本研究ではアサガオを材料にして、これらの過程に関わる遺伝子などを単離・同定して花色との関連を解明し、液胞の機能と役割について総合的に明らかにすることを目的としている。本年度は下記のような知見を得た。
1)アントシアニン色素にグルコースを付加する新規の酵素遺伝子をクローン化した。また、花色が暗色化するdusky変異は、本酵素遺伝子の4bpの挿入変異であることを見いだした。さらに、本酵素遺伝子を導入したペチュニアで、ペチュニアには存在しないアントシアニンの生合成と花色の変化を確認した。
2)アサガオ花弁の表層細胞のpHは開花に伴い一過的に6.6から7.7程度に上昇し、昨年度までに、この一過的上昇に液胞型Na^+/H^+exchenger(NHX1)が関わることを明らかにしている。本年度はNHXと同調して働くことでpH上昇に関わる遺伝子の探索を行い、サブトラクション法やESTデーターベースの検索により侯補遺伝子を複数得ることに成功した。一方、NHX1に相同性が高いNHX2遺伝子もクローン化して発現を検討したが、NHX1とは異なり花弁での発現は僅かで実生にナトリウムを与えることで誘導されることから、花色ではなく対塩性に関わることが示唆された。
3)トウモロコシおよびペチュニアでアシトシアニンの液胞への輸送に関わるとされるグルタチオンS-転移酵素(GST)遺伝子の相同遣伝子をクローン化した。色素生合成系遺伝子の転写を活性化する遺伝子の支配下にあることから、アサガオにおいてもGSTが色素の輸送に関わることが示唆された。
