Research Abstract

神経予定外胚葉はまず中枢神経系原基である神経板と末梢神経原基である神経堤細胞とに分画化される。中枢神経原基・神経板は発生のごく初期に吻尾方向と背腹方向の2軸に沿って大きく分画化され、いわゆる領域特異性を獲得する。吻尾方向には大脳・間脳、中脳、後脳、脊髄が大きく区分され、背腹軸では背側(翼板)、腹側(基板)、中間部に区分される。それぞれの領域には特異的な分子マーカー(ホメオボックス遺伝子など)が既に同定されており、それらを用いて神経細胞がどの領域特異性を獲得したかを判定することが原則的に可能である。しかし、この領域特異性の上流にあって、その個性付け獲得を制御している因子については多くが不明のままである。そこで、領域特異性の上流にある神経分化の個性付け因子を系統的に遺伝子スクリーニングすることを行った。まず初期神経板で働く領域特異的分泌タンパクを系統的にシグナル・シーケンス・トラップ法によって用いて、アフリカツメガエルの系で神経管の背側に位置する非神経外胚葉に早期から発現する新規の分泌因子Tiarinを単離に成功した。H15年度は単離したマウスおよびニワトリホモローグを用いて、これちの種での機能について強制発現を用いて解析し、神経提細胞の産生促進効果を観察した。また、現在2種類のマウス関連遺伝子に関して遺伝子破壊法で機能阻害研究を進めている。研究の促進のため、ES細胞から神経前駆細胞を分化させ、これを用いた試験管内神経パターン形成のアッセイ系を確立した。この系を用いて末梢神経系を含む神経提細胞のES細胞からの分化に世界で初めて成功した。
