Research Abstract

運動ニューロンのidentityを反映する分子的実体を明らかにするため、我々は分泌あるいは膜結合型蛋白質に存在するシグナルシークエンスを持ち、且つ運動ニューロンサブタイプに選択的に発現されるcDNAを探索するためシグナルシークエンストラップ法を遂行した。
その結果、発生初期胚の運動ニューロンに選択的に発現されている新規膜蛋白質10B4と273分子を見出した。これらの分子には、分子構造として蛋白質間の結合活性を持つロイシンリッチリピートが細胞外領域に存在するため、その細胞外領域とアルカリフォスファターゼとの融合蛋白質を作成し、ニワトリ胚での結合相手を探索したが選択的に結合する組織や細胞は見いだせず、両分子共に可溶性分子と結合する受容体としての機能を持つと推察された。実際、273は細胞質内領域のチロシン残基がリン酸化されていた。モノクローナル抗体を作製した免疫組織化学の解析から、273分子は運動ニューロン軸索が伸展し始める時期には細胞体と軸索双方に発現していたが、筋との結合が形成され始めるE6以降では細胞体の発現は消失していき、軸索には孵化するE21近くまで発現が持続した。これらのことから、筋やシュワン細胞からの何らかの因子を受容しているのではないかと想定された。
これらの分子の機能を明らかにするために、電気穿孔法によるニワトリ胚内への遺伝子導入による過剰発現やsiRNAによる機能阻害を試みてきたが、これまでのところ、明確な機能を見いだせていない。しかし、10B4分子は2004年3月号のNature NeuroscienceにNogo-66/p75受容体コンプレックスと協同的に働く分子として報告されたLINGO-1と同じ分子であった。今後は、273と10B4分子の発生過程における機能を解明していく研究を継続する予定である。
