Research Abstract |
Trace clements such as Se and Zn are involved in redox regulation by comprising antioxidative enzymes or proteins. Metallothionein, a Zn-binding protein ubiquitously expressed in the body, was shown to affect the activation of NF-kB, a stress-inducible transcription factor when cells were stimulated by TNF-α. On the other hand, thioredoxin reductase (TrxR), an enzyme responsible for the maintenance of reduced form of thioredoxin, was found to be a Se-containing enzyme. In bovine arterial endothelial cells (BAEC) cultured in Se-deficient medium, the activity of TrxR was reduced while the mRNA level of TrxRl (cytosolic form of TrxR) was increased. Treatment of BAEC with hydrogen peroxide resulted in an increase in synthesis of TrxRl as judged by mRNA level and Se incorporation, while no changes were observed in other selenoproteins. These results suggest that the regulation of TrxR is differently regulated by Se nutrition and oxidative stress.
