Research Abstract |
Functional recovery in brain damages is fundamentally caused by the modulation of synaptic transmission in the mature brain. By using positron CT, we have developed the method in which external measurement is possible in order to know this modulation function in the human brain. In this report, higher brain function recovery mechanism after the brain damage was examined using this method for the relation between enhancement phenomenon of the inositol phospholipid signal and an improvement of higher brain function. The phenomenon appeared in probability (75%) in the brain damage case in the subacute stage in central nervous system injury or cerebral apoplexy. It was proven that it did not appear chronic stage. That is to say, it is possible to regard as this interval from 2 weeks to 1 month in the repair stage. and improvement on the memory function. In the Alzheimer disease case, radioactive spot which shows high integration of the tracer in the frontal association area was also recognized. It was indicated that some plastic changes existed for the frontal association area of the Alzheimer disease patient. Confocal laser microscope and fluorescent labeling DAG were newly synthesized. As a result of examining the localization of the enhancement phenomenon by the micro level, it was proven that it occurred in the dendrite. The appearance part is confirmed by the serial section of the rat brain, and this position is taken out for differential display method. The factor which was concerned in the modulation of the synaptic transmission using the method was searched. However, the significant difference in the reference could not be recognized. Probably in the stage in the alteration of the phospholipid membrane structure, it seemed to be not generating the change by the mRNA level. The dyeing agreed with the position of the enhancement phenomenon in the immunohistochemical technique using the CaMKII-active antibody, and the relevance to the Ca ion current was indicated. Fluorescent labeling DAG and positron-labeled DAG seemed to accumulate by the degeneration of the phospholipid of the cell membrane induced from this fact by the Ca ion current.
