Research Abstract |
1 Correlation between Bax, bcl-2 protein expression and radiation-induced apoptosis Bax bcl-2 protein expression correlate with radiation-induced apoptosis on 21 cases with the cervical cancer, including 11 cases of stage 2, 8 cases of stage 3, and 2 cases of stage 4 was investigated. Apoptosis was detected by using Apoptag (Oncor), and Apoptotic Index (AI) was calculated. Bax and bcl-2 protein expressions were detected with both anti-Bax antibody (Oncor) and anti-bcl-2 antibody (DAKO)immunohistochemically. RESULT : Apoptosis was 0.02% before irradiation, and it significantly increased up to 1.2% at 9Gy. The Dositive rate of Bax was 15% before irradiation, and it increased up to 60% at 9Gy. While that of bcl-2 was 15% before irradiation, and it increased to 25% at 9Gy which was no significant increase. Additionally, a significant correlation was observed between Bax expression and apoptosis of the tumor at 9Gy. CONCLUSION : Bax protein expression significantly correlates with radiation-induced apoptosis of cervical cancer. 2 Correlation between p53, p21 protein expression and radiation-induced apoptosis Correlation between p53, p21 tumor expression and radiation-induced apoptosis were investigated. Before irradiation, the mean AI of p53 and p21 were 6.60% and 9.24% respectively. Then after 9Gy the mean AI of p53 and p21 significantly increased to 13.9% and 18.0% respectively. A significant positive correlation appeared between p53 protein index and p21 protein index before irradiation. Mean AI, p53 index, and p21 index were 0.98%, 7.33% and 8.77% respectively. As for radiation-induced AI, a significant negative correlation was observed between both AI and p53 or p21. Moreover, a significant positive correlation was observed between both p53 and p21. CONCLOSION : The cascade of p53 and p21 act as apoptosis inhibitory action in cervical cancer.
