Research Abstract |
1. Tolerance induction of pancreatic allograft by intrathymic islet transplantation : F344 rat islets were transplanted into the thymus of diabeetic WKAH rat with single injection of ALS. Three weeks after intrathymic transplantation, pancreatic graft of F344 rat was transplanted into WKAH rat. Pancreatig graft survival was sfive days (Median survival), which did not show the prolongation as compared to control (6 days). 2. Development of Chiba Univ. Islet Preparation Method : We designed a new islet preparation system using mechanical chopper, automated 2-step digestion unit, HES purification with COBE2991 cell professor. Using Beagle dogs, significant improvement of islet yield, purity, insulin release were obtained with our new method as compared to the conventional method. 3. Islet isolation from non-heart beating donor : Using Beagle dogs, pancreas were harvested with 30 minutes' warm ischemia with or without in situ machine wash out. High quality islet were isolated when using in situ machine wash out.
