Research Abstract |
In order to investigate whether proton pump inhibitor (PPI) ameliorates glucose tolerance after pancreatectomy through hypergastrinemia, 90% pancreatectomized rats (male Wistar) were treated as follows ; Group 1 : vehicle alone, Group 2 : PPI (lansoprazol, 30 mg/kg body weigh), Group 3 : gastrin/CCK-B receptor inhibitor (GRI, AR-041R, 30 mg/kg BW), Group 4 : PPI and GRI. The drugs were daily administered into the stomach. Normal rats were also examined. Fourteen days after pancreatectomy (Px), plasma gastrin levels were significantly increased by PPI and tended to be additionally increased by GRI. Wet weight of the organ/body weight ratio (ww/BW) of the pabcreas was increased in Px rats in comparison to the remnant equivalent of the normal rats. However, significant difference was not seen among the Px groups. The ww/bw of the stomach was significantly increased in Px groups and partly reduced by GRI. Either changes in body weight or plasma glucose concentration at fed state were not different among Px groups. Intragastric glucose challenge (2g/kg BW) did not show the difference in plasma glucose or insulin concentrations among Px groups. This study did not reproduce the effect of PPI on glucose tolerance after Px, however the changes in gastric weight was reproductively observed as was shown in our previous study. ィイD1*ィエD1Cahnges in glucose tolerance after Px seemed to be delicately influenced by a little differences in the rate of Px and/or other unknown factors. On the other hand, the role of gastrin on Px-induced increase in gastric weight was partly evaluated by the effect of GRI, which fully antagonized PPI-induced increase but did not abolish the Px-induced increase. Therefore, some factor(s) other than circulation gastrin mainly mediates Px-induced increase in gastric weight. ィイD1*ィエD1 : Xu G, Sumi S, et al.
