Research Abstract |
We performed electron microscopic studies of 11 non-functioning adrenocortical adenomas (NFA), 2 adenomas with pre-clinical Cushing's syndrome, 10 aldosterone-producing adenomas (APA) and 4 adenomas with Cushing's syndrome, obtained by surgical extirpation. Ultrastructurally, 9 of 11 NFA were classified into the aldosteron-producing type and 2 into the Cushing's type. The ultrastructural features of adenomas with pre-clinical Cushing's syndrome were similar those of adenomas with Cushing's syndrome. Based on morphometric analysis, the mean ratio and standard deviation (SD) of mitochondria (M), smooth-surface endoplasmic reticulum (SER), rough-surface endoplasmic reticulum (RER), lipid vacules (LV) and lysosomes (Ly) was 11.2 ± 7.7%, 16.3 ± 13.8%, 2.8 ± 2.0%, 20.0 ± 23.5% and 1.3 ± 1.9% in the NFA, and 17.2 ± 8.6%, 19.3 ± 9.8%, 6.3 ± 5.5%, 19.4 ± 13.1% and 1.2 ± 1.6% in the APA, respectively. The ratio of M (p<0.01), SER (p<0.01) and RER (p<0.01) in the NFA significantly declined compared to that in the APA. The ratio of LV in NFA resembling adenomas with Cushing's syndrome and adenomas with pre-clinical Cushing's syndrome significantly increased compared to that of adenomas with Cushing's syndrome. The ratio of M and SER in the NFA resembling adenomas with Cushing's syndrome significantly decreased compared to that in adenomas with Cushing's syndrome, whereas the ratios of these organelles between adenomas with Pre-Cushing syndrome and those with Cushing's syndrome were not statistically different. The difference of p-450scc localization among NFA, APA and adenomas with Cushing's syndrome was not found based on immuno-electron microscopic study.
