Research Abstract |
Inhibin dimmer inhibits FSH secretion from anterior pituitary cells in primary culture and inhibin α-monomer has no effect on FSH secretion. But as reported so far, the action of inhibin is different from tissue to tissue. In order to establish a suitable system to investigate the action of inhibin, we investigated several cells using activin because the availability of inhibin is very limited and inhibin and activin use common receptors. (1) Pituitary cells : The regulation of PRL secretion by activin from a rat pituitary cell line with type-I and type-II activin receptor complex on the cell surface was investigated. PRL secretion was changed by activin. Further study regarding the involvement of the inhibin/activin signal in PRL production is useful for our study. (2) Ovarian cells : Granulosa cells from DES-primed immature rats and oocyte-granulosa-theca cell complex from immature mice reacted to activin in low concentrations. Therefore these cells seem to be suitable in order to study the action of inhibin dimmer and α-monomer. (3) Placental cells : A primary culture of human placental cells at term was investigated. Only high concentrations of activin changed the secretion of hCG so these cells do not seem suitable to use to investigate the local action of inhibin.
